Her best-known role is her roles in Spamalot's West End revival, the Regent's Park revival of Into the Woods, as well as The Wizard of Oz (as the Wicked Witch of the West). Her work has been nominated three time for the Olivier Award. In 2015, she joined the Game of Thrones cast as Septa Unella The sixth season of the fifth season. In addition to her main role as Septa Unella as well, she was also an ensemble member of the 2012 film adaptation of Les Miserables as well as the 2018 British psychological thriller Winter Ridge. Waddingham was nominated for the Golden Globe Award. She has been playing the character of Rebecca Welton on the Apple TV+ comedy series Ted Lasso from 2020.
